利用低温真空干燥技术对棉纤维起始伸长的扫描电镜观察

摘要: 棉纤维由棉花受精胚珠的表皮细胞伸长加厚而成,其起始伸长过程是棉纤维发育的重要阶段。为了更好地观察棉纤维在发育过程中的形态变化,利用低温真空干燥技术,对棉花胚珠表皮细胞突起和伸长过程进行扫描电镜观察,获得了理想的观察结果。通过与传统的叔丁醇乙腈干燥法处理样品的扫描电镜图片进行比较,利用低温真空干燥技术处理的样品形态饱满,轮廓清晰,能真实地观察样品形态。所采用的方法为扫描电镜的使用提供了一个典型范例,对其它植物或含水量较高的生物样品的制样和观察实验具有重要的参考价值。Abstract: Cotton fibers are single‐celled trichomes that emerge from the epidermal cells of fertilized ovule . In order to study the development of the cotton fiber ,we use the Cryo‐SEM with high vacuum to observe the initiation and elongation of ovule epidermal cells and the good results have been observed .Compared with the tert‐butyl alcohol vacuum‐drying method ,the results of observations are more chubbiness and clearer outline by our methods .This method will provide a typical example of material preparation for scanning electron microscope ,w hich has important influence for the SEM observation in other plants or bi‐ological samples with high water content .
