玉米几丁质酶基因导入甘蓝型油菜的研究

摘要: 以甘蓝型油莱(Brassica napus L.)下胚轴为转化材料,通过根癌农杆茵(Agrobacterium tumefaciens)介导将玉米几丁质酶基因(Maize Chitinsse gene)导入双低油菜品种D2中,获得了抗卡那霉素(Kanamycin Kan)的再生植株,并对影响遗传转化的一些因素进行了研究。结果表明:适当的预培养对油菜下胚轴的遗传转化是必要的;油菜下胚轴遗传转化过程中的过敏反应可通过一些措施缓解;延迟筛选可提高油菜下胚轴的转化率。对抗性植株进行PCR检测表明:部分抗性植株在多次重复检测中均显示较强的阳性反应,初步证明外源基因已整合到油菜基因组中。Abstract: In an experiment with the hypocotyls of rapeseed (Brassica napus L.) as explants in in vitro culture, a transformation of introducing maize chitinase gene into the double- low rapeseed cultivar D2 by Agrobacterium tumefaciens was established. Some Kan- resistant green shoots were obtained. It was shown that pre- eulture was essential for the transformation of the hypocotyls ofrapeseed,the hypersensitive responses of the cultured hypocotyls in transfornation could be alleviated by some measures, and delayed selection was advantageous to increasing transformation frequency. PCR test ofthe resistant plants indicated that some ff the Kan- resistant plantlets showed strong positive reaction in repeated tests,suggesting that the foreign gene had been integrated into the genome of rapeseed.
