摘要: 用随机扩增多态性DNA(RAPD)分析技术对甘薯及其近缘野生种进行了遗传多样性分析.使用15种10个碱基的随机引物,对17个甘薯品种及其近缘野生种的基因组DNA进行了扩增,共产生了105条DNA带,其中91条为多态性带,通过聚类分析将17个甘薯品种及其近缘野生种聚类成3个类群:A类群由三裂叶野牵牛(Ipomoea triloba)单个野生种组成,C类群由海滨野牵牛(I.littoralis)和二倍体三浅裂野牵牛(I.trifida2x)组成,B类群则由四倍体和六倍体三浅裂野牵牛以及12个甘薯品种组成.

Abstract: The genetic relationships among 17 swe et potato (Ipomoea batatas (L.)Lam.)cultivars and related wild species—I . tril oba (2n=2x=30), I. littoralis (2n=4x=60) and I. trifida (2n=2x=30, 2n=4x=60, 2n=6x=90) were studied with RAPD (Random Amplified Polymorphic DNA) analysis that based on PCR(Polymerase Chain Reaction). Fifteen 10—mer primers were scree ned from sixty arbitrary primers. A total of 105 DNA bands were amplified, among them, 91 bands were polymorphic ones which accouted for 86.67 percent of the t otal. the clustering dendrogram with coefficent of Euclidean distance clustered. The result was classified into three cluster groups. Only one wild species— I. triloba,was in group A, and two species—I. littoralis and I.trifida( 2x), were in group C. The group B consisted of tetraploid and hexaploid wild s pecies (I. trifida 4x, 6x) and 12 sweet potato cultivars. -
