鸦胆子的组织培养与快速繁殖

摘要: 以鸦胆子顶芽、侧芽为材料,采用正交实验方法,研究鸦胆子组培快繁技术.结果表明,①组合 MS+NAA 0.5 mg/L+KT 1.0 mg/L+6 BA 2.0 mg/L有利于鸦胆子不定芽的诱导,诱导率为100%,不定芽数量为2.725个;②组合6 BA 1.0 mg/L+NAA 0.05 mg/L+KT 1.0 mg/L+40 g/L蔗糖不定芽增殖率最高(2.75),同时高质量浓度(1.0 mg/L)KT处理有利于不定芽增殖,低质量浓度(20 g/L)的蔗糖不利于不定芽增殖;③1/2 MS+(0.5~1.0) mg/L IBA(或NAA)均能诱导不定芽生根,生根率为89.3%.Abstract: Terminal and lateral bud of Brucea j av anica (Linn) Merr has been selected as explants to carry out the plant tissue culture technology research by means of the orthogonal experiment .The results show that plant hormone combination MS+NAA 0 .5 mg/L+KT 1 .0 mg/L+6‐BA 2 .0 mg/L is good for adven‐titious buds induction ,and induction rate is 100% ,numbers of adventitious buds are 2 .725 .Plant hor‐mone combination 6‐BA 1 .0 mg/L+NAA 0 .05 mg/L+KT 1 .0 mg/L+40 g/L sucrose shows the highest propagation rate (2 .75) for adventitious buds ,and high concentration of KT treatment (1 .0 mg/L) is fa‐vorable for adventitious buds propagation but low sucrose concentration treatment (20 g/L )is unfavorable for adventitious buds proliferation .1/2 MS+ (0 .5~1 .0)mg/L IBA(or NAA)can induce adventitious buds to take root and rooting rate was 89 .3% .
