荧光素-GOD-HRP 体系分光光度法测定植物 果实组织中的葡萄糖

摘要: 植物组织中的葡萄糖在葡萄糖氧化酶(GOD )作用下产生的过氧化氢可在辣根过氧化物酶(HRP)的催化作用下使荧光素产生褪色反应,基于这一现象建立了一种选择性测定植物果实组织中葡萄糖含量的方法. 该方法可以根据检测对象的不同调整检测范围和灵敏度,同时具备了安全,方便的优点.利用此法对苹果、鸭梨、酥梨中的葡萄糖含量进行了测定,结果与化学发光法一致.Abstract: A new spectrophotometric method for the determin ation of glucose based on the fading of fluorescein in the presence of glucose, glucose oxidase and horseradish peroxidase was proposed.The method is selective and relatively sensitive, especially suitable to analysis of glucose in analyte containing sucrose and fructose. The method could be linear in 0~50 μg/mL range for glucose and obeyed the following equation: ΔA = 0.007 3C + 0.005 3 (n=8, r=0.992 7). In this work, glucose content in apple, Sinkiang pear and Hopei pear were determined and the result agreed well with those obtai ned from chemiluminescence method.
